A pure substance is composed of same kind of particles and a mixture of different kind of particles.pure substance have definite properties i.e.fixed melting and boiling points.   But mixtures have the properties of constituents of which they are composed of. .... P.S are always homogeneous but mixtures can be homogenous or hetrogeneous.

One way to tell the difference is by first measuring the density. This will help you to determine which substance is what by seeing which two substances will have an average equal to the density of another substance. Another way is to boil/melt the substance. The pure substance will have one set melting point while the mixture will have two or more different melting points.
